Deacetoxycephalosporin C synthase

(DAOCS)



Penicillins and cephalosporins are antibiotics that are produced from fermentation derived material through biosynthesis. Deacetoxycephalosporin C synthase (DAOCS) catalyses the expansion of the penicillin nucleus into the nucleus of cephalosporins. The reaction requires dioxygen and 2-oxoglutarate.


We have determined the crystal structures for the prokaryotic DAOCS from Streptomyces clavuligerus: the iron-free apoenzyme, the enzyme complexed with Fe(II), and the enzyme complexed with Fe(II) and 2-oxoglutarate.
